Assuming you mean layouts, yes, it can work. But its generally better practice to develop page templates for any layout you are using in more than one place or page-wide. Page templates are more robust and as long as you can do a little php, easier to maintain and manage.

As a rule of thumb, layouts are something to use for one off situations where you need a split column for a specific small part of a page.
