Missing index page

We have a C5 site that seems to have had the index page go missing. If you visithttp://www.spcai.org you get a 404 but if you usehttp://www.spcai.org/index.php it works fine. As you would expect clicking home in the nav comes up with the same 404.
We used some URL redirects with URL Director but even turning them off we have the same, seemingly mysterious error.
Any ideas? And thank you very much in advance.
