Hi, I'm building quite a large and complex site in C5.
I have the home page then 5 sub-home pages each with a different colour theme and then the detail pages in which will have most of the content.
Should I build the sub-home pages as single pages (5 of them as they will only be used once) then then have my detail pages set up as page types.
Or does this not really matter if you use the single pages or page types?

If I were you, I would use 'Page Types' rather than 'Single Pages' for your 5 sub-nodes pages.
Now you might be 100% sure that each of these sub-pages will be used only once, but what if in a couple of months you or your client decide to use one of this sub-pages in other part of the website?
This kind of things happen all the time.
Also, as far as I know, there is no extra benefits in using them as Single Pages.
