Understanding DIV's + Responsive layouts

Hi all,
I am trying to understand a very basic concept which I have realised that I can't yet grasp. I'd like to know if the task I have set myself is an easy one to learn, or best left until later in my learning curve (see attachment).
I created a concept for a client and have been slowly modifying the Slate theme, before realising that I don't quite understand how DIV's shuffle around, or the concept of rows!
Inside the CSS of Chris' package is this area referring to "Columnal - sub-columns". This led me tohttp://www.columnal.com/
Now I am trying to undertand how sub-columns work with DIV's and rows.
I have attached a file with a sketch showing a conceptual layout, and am wondering if this is worth pursuing as a responsive site, or if this is something that is considered to be rather difficult by more seasoned web developers.
