Major Issues!!!

Hi I have a bit of an emergency with major issues with at least two websites.
See first indication error in screenshot.
From there you get a lot of Page not found errors, see second screen shot.
Then the site basically goes haywire.
Clearing the cache does alter it. The site starts to behave but not completely. Turning the cache off and you cant get access at all. It seems to be a cache issue and only since upgrade to 5.4.1.

Seems to be worse with pages with "layouts" added.
